Cellular Cycling, Carbon Utilization, and Photosynthetic Oxygen Production during Bicarbonate-Induced Triacylglycerol Accumulation in a Scenedesmus sp

چکیده
Microalgae are capable of synthesizing high levels of triacylglycerol (TAG) which can be used as precursor compounds for fuels and specialty chemicals. Algal TAG accumulation typically occurs when cellular cycling is delayed or arrested due to nutrient limitation, an environmental challenge (e.g., pH, light, temperature stress), or by chemical addition. This work is a continuation of previous studies detailing sodium bicarbonate-induced TAG accumulation in the alkaline chlorophyte Scenedesmus sp. WC-1. It was found that upon sodium bicarbonate amendment, bicarbonate is the ion responsible for TAG accumulation; a culture amendment of approximately 15 mM bicarbonate was sufficient to arrest the cellular cycle and switch the algal metabolism from high growth to a TAG accumulating state. However, the cultures were limited in dissolved inorganic carbon one day after the amendment, suggesting additional carbon supplementation was necessary. Therefore, additional abiotic and biotic experimentation was performed to evaluate inand out-gassing of CO2. Cultures to which 40–50 mM of sodium bicarbonate were added consumed DIC faster than CO2 could ingas during the light hours and total photosynthetic OPEN ACCESS Energies 2013, 6 6061 oxygen production was elevated as compared to cultures that did not receive supplemental inorganic carbon.

A series of experiments were carried out with three native strains of microalgae to measure growth rates, biomass, and lipid productivities. Scenedesmus sp. IMMTCC-6 had better biomass growth rate and higher lipid production. The growth, lipid accumulation, and carbon dioxide (CO2) consumption rate of Scenedesmus sp. IMMTCC-6 were tested under different NaOH concentrations in modified BBM. The ...
متن کاملSuperior triacylglycerol (TAG) accumulation in starchless mutants of Scenedesmus obliquus: (II) evaluation of TAG yield and productivity in controlled photobioreactors
BACKGROUND Many microalgae accumulate carbohydrates simultaneously with triacylglycerol (TAG) upon nitrogen starvation, and these products compete for photosynthetic products and metabolites from the central carbon metabolism. As shown for starchless mutants of the non-oleaginous model alga Chlamydomonas reinhardtii, reduced carbohydrate synthesis can enhance TAG production. However, these muta...

The freshwater green alga Scenedesmus opoliensis proves to be a suitable bioindicator of water pollution with different herbicides. One of the best molecular markers of stress condition imposed by herbicides is overproduction of malondialdehyde resulting from lipid peroxidation in the damaged membranes. Methylviologen, a largely used pre-emergence herbicide which generates reactive oxygen speci...
متن کاملTwo Systems for Concentrating CO(2) and Bicarbonate during Photosynthesis by Scenedesmus.
Scenedesmus cells grown on high CO(2), when adapted to air levels of CO(2) for 4 to 6 hours in the light, formed two concentrating processes for dissolved inorganic carbon: one for utilizing CO(2) from medium of pH 5 to 8 and one for bicarbonate accumulation from medium of pH 7 to 11. Similar results were obtained with assays by photosynthetic O(2) evolution or by accumulation of dissolved inor...

Microalgae are a large and diverse group of photosynthetic organisms which can use of solar energy to fixed inorganic carbon (Ci) with efficiency (2-10%) that are ten times greater than higher plants (<1%) due to their high growth rate (1to 3-fold increases in biomass per day) [1,2]. Although CO2 is the predominant carbon source for microalgal cultivation, the low level of dissolution of CO2 in...
